1）细胞培养物上清：细胞培养物于室温1000g，离心10分钟后收集上清，并将标本保存于-20℃，且应避免反复冻融。
2）血清：标本请于室温放置2小时或4℃过夜后于1000g，4℃离心20分钟，取上清即可检测，或将标本放于-20℃保存，但应避免反复冻融。
3）血浆：可用EDTA或肝素作为抗凝剂标本采集后30分钟内于2-8℃,1000g离心15分钟，或将标本放于-20℃保存，但应避免反复冻融。
4）尿液：无菌收集取初尿，离心除去沉淀物，即可用于检测，要长期保存尿样，可以加入0.05％的NaN3在4-8℃保存，或加入尿样冰冻保护液放入-20℃冰箱长期保存。
5）组织匀浆：将组织标本先用PBS洗涤，除去多余血液，匀浆化后放在PBS于-20℃放置过夜，再经过二次反复冻融破膜，5000g，4℃离心5分钟，取上清即可检测。
注：取样和样品处理过程中，防止有毒物质对操作人员的危害，要采取相应的保护措施。

ELISA定性测定结果判定中常用的一些缩写：
（1）S／CO：其中S为sample（样本）或specimen（标本）的简写，表示的是标本测定的吸光度值，CO为cut-off值的简写。除竞争抑制法外，其他ELISA定性测定模式中，当S／CO值大于或等于1时，标本的测定为阳性，小于1时为阴性。
（2）S／N或P／N：其中S同（1），N为negative（阴性对照）的简写，P为patient（患者）的简写。较早的试剂盒很多都使用S／N或P／N≥2．1为阳性判定标准，现仍有一些人β干扰素（IFN-β/IFNB）ELISA试剂盒，48T/96T使用这种方式。这种方式与S／CO方式无根本性区别，只不过是前者将阴性对照（N）的2．1倍视为cut-off值而已。
